Monoklonale Antikörper gewinnen aufgrund ihrer vielseitigen Anwendungsbereiche immer weiter an Relevanz. Sie werden sowohl in der Diagnostik als auch in therapeutischen Maßnahmen eingesetzt und machen bereits den Großteil der biotechnologisch produzierten Biopharmaka aus. Um die Herstellungsprozesse neuer Antikörper systematisch hinsichtlich der Prozess- und Produktqualität zu verbessern, soll der Quality by Design Ansatz in die biopharmazeutische Forschung und Industrie integriert werden. Dabei mangelt es jedoch weiterhin an Modellprozessen für die praktische Implementierung über eine ganzheitliche Prozesskette. In der vorliegenden Dissertation wurde der Upstream-Prozess in einzelne Prozessschritte eingeteilt und anhand der Quality by Design Prinzipien charakterisiert. Dafür wurden zunächst kritische Prozessparameter und Qualitätsattribute der einzelnen Prozessschritte identifiziert. Anschließend wurden die Parametereffekte- und Interaktionen mithilfe von statistischen Versuchsplänen analysiert und ein robuster Design Space etabliert.
Im ersten Teil wurde die Vorkultur als kritischer Teil des Herstellungsprozesses eingestuft. Dabei wurden die Einflüsse der Parameter auf Qualitätsattribute der Vorkultur und des nachfolgenden Prozesses analysiert. Als Schlüsselparameter wurden die Passagendauer und die viable Zelldichte der Passagen identifiziert.
Während der Quality by Design Integration in den Produktionsprozess wurden zunächst prozessbezogene Qualitätsattribute untersucht, wobei der pH-Wert des Kulturmediums und die initiale Zelldichte als kritische Parameter festgestellt wurden. Die Bioaktivität des produzierten Antikörpers zeigte innerhalb des charakterisierten Bereiches jedoch keine signifikanten Veränderungen.
Im dritten Teil wurde die Zellabtrennung als Bindeglied zum Downstream-Prozess analysiert. Die Viabilität wurde als Schlüsselparameter mit großem Einfluss auf prozessbezogene Verunreinigungen identifiziert. Die etablierten Design Spaces definierten hohes Zellwachstum und Zellerhaltung bei optimaler Antikörperproduktion als Ziel der Prozessführung und -Kontrolle.
Monoclonal antibodies are becoming increasingly relevant due to their versatile areas of application. They are used in diagnostics as well as in therapeutic measures and already account for the majority of biotechnologically produced therapeutics. In order to systematically improve the manufacturing processes of new therapeutic antibodies in terms of process and product quality, the Quality by Design approach is to be integrated into biopharmaceutical research and industry. However, there is still a lack of model processes for practical implementation across a holistic process chain. In this dissertation, the manufacturing process of a monoclonal antibody was characterized. The focus was on the application of quality by design principles in different sub-steps of the upstream process.
In the first part, the inoculum expansion was studied and classified as a critical part of the production process. The influences of the parameters on quality attributes of the preculture and the subsequent production process were analyzed. Passenger duration and viable cell density of passages were identified as key parameters. The second part describes the Quality by Design integration into the production process. Process-related quality attributes were first investigated, with the pH of the culture medium and the initial cell density found to be critical parameters. In contrast, the bioactivity of the produced antibody showed no significant changes within the characterized range.
In the third part, cell separation was analyzed as a link to the downstream process. The viability was identified as a key parameter with major influence on process-related impurities. The established design spaces defined high cell growth and cell maintenance with optimal antibody production as the goal of process analysis and control.
