Im Bereich der Bioprozessentwicklung sind Miniaturisierung, Parallelisierung und Flexibilität entscheidend, um Zeit und Kosten zu sparen und Ressourcen zu schonen. Um genau diese Anforderungen zu erfüllen, ist die additive Fertigung (3D-Druck) eine ideale Technologie. Der 3D-Druck ermöglicht eine schnelle Herstellung von Prototypen und eine kostengünstige Produktion von maßgeschneiderten Komponenten mit komplexen Geometrien nach Bedarf.
Im Rahmen dieser Arbeit wurde ein 3D-gedrucktes Kultivierungsgefäß entwickelt, welches mittels Inkjetverfahren aus einem transluzenten Polyacrylat gefertigt wurde und ein Gesamtvolumen von ca. 50 mL aufweist. Es wurde so konzipiert, dass es mit dem Shake Flask Reader vario (SFR vario), einem Multisensorsystem für Schüttelkolben, kompatibel ist. Dafür erfolgte die Integration von optischen Sensoren für die Online-Überwachung von pH und Gelöstsauerstoffkonzentration. Die stetige Weiterentwicklung und Optimierung des ersten Prototypens während der Charakterisierungsexperimente führten zu dem optisch modifizierten Well (OMW). Durch eine veränderte Lichtführung mittels 3D-gedruckter Prismen konnte die streulichtbasierte Biomassemessung mit Hilfe des SFR varios deutlich verbessert werden. Die Verwendung des OMWs für Upstream-Prozesse verschiedener Mikroorganismen und Zelllinien wurde in dieser Arbeit evaluiert. Es wurde gezeigt, dass das OMW für Kultivierungsvorhaben mit Escherichia coli, Bacillus subtilis, Saccharomyces cerevisiae und Chinese hamster ovary (CHO) Zellen geeignet ist. Es liefert reproduzierbare Ergebnisse und ermöglicht die präzise Überwachung von Kultivierungsphasen mit geringen optischen Dichten unter 1,0 rel.AU. Bei gleichem Wachstumsverhalten der OMW-Kulturen im Vergleich mit Schüttelkolbenkulturen konnte das Signal-Rausch-Verhältnis signifikant reduziert werden.
Zur Validierung des 3D gedruckten OMWs wurde die Übertragbarkeit von drei Bioprozessen zur Herstellung heterologer Produkte auf untersucht: die Produktion einer Carboxylsäurereduktase in E. coli, die ganzzellkatalytische Produktion des Sesquiterpens Germacren A in E. coli und die Produktion des grün fluoreszierenden Proteins (GFP) in E. coli. Dabei wurden die Vorteile des 3D-Drucks genutzt und das Standard-OMW umdesignt, sodass ein weiterer kommerzieller Sensor zur Online-Überwachung der Fluoreszenz integriert werden konnte.
In the field of bioprocess development the concept of miniaturization, parallelization and flexibility are pivotal in the effort reducing time and costs and saving valuable resources. To meet precisely these requirements, additive manufacturing (3D printing) is an ideal technology. 3D printing facilitates the rapid prototyping and the cost-effective production of customized components with complex geometries on demand. In this work, a 3D-printed cultivation vessel was developed, manufactured from a translucent polyacrylate material, using the multi jet modeling technology and holding a total volume of approx. 50 mL. It was designed to be compatible with the multi-sensor system Shake Flask Reader vario (SFR vario) for shake flasks. For this purpose, optical sensors were integrated for online monitoring of pH and DO. Continuous development and optimization of the first prototype during the characterization experiments led to the optically modified well (OMW). By modifying the light guidance using 3D-printed prisms, it was possible to significantly improve the scattered light-based biomass measurement using the SFR vario. In this work, the use of the OMW for upstream processes of various industrially relevant microorganisms and cell lines was evaluated. It was shown that the OMW is suitable for cultivation purposes with Escherichia coli, Bacillus subtilis, Saccharomyces cerevisiae and Chinese hamster ovary (CHO) cells. It provides highly reproducible results and allows precise monitoring of cultivation phases with low optical densities below 1.0 rel.AU. With the same growth behavior of OMW cultures compared to shake flask cultures, the signal-to-noise ratio could be reduced by up to 96 %. Furthermore, it was shown that the 3D-printed optics are transferable to different vessel geometries and sizes, enabling efficient biomass monitoring for individual requirements with customized 3D-printed cultivation vessels on a small scale.
In addition, the transferability of bioprocesses for the production of heterologous products to the 3D-printed OMW was investigated: the production of a carboxylic acid reductase in E. coli, the whole-cell catalytic production of the sesquiterpene germacrene A in E. coli and the production of green fluorescent protein (GFP). Here, the advantages of 3D printing were used and the standard OMW was redesigned in such a way that an additional commercial sensor could be integrated for online monitoring of the fluorescence.
